Hi, strange lock issue here. its a 98 4runner, sr5.
Just noticed today that when i put my truck into drive, all the doors locked, except my drivers side. When I got where I was going and attempted to use the power lock, all doors locked except the drivers side. Locked it manually. when I tried to use the key in the door, the lock will not turn, left or right. The lock functions, as in I can move the post up and down manually, and it will lock and unlock in that way only. I am confused by why the key wont manually turn the lock. Any thoughts?

I have a problem like this on my BMW, if your key is double sided try both sides by turning the key over.

 

its not frozen is it? I'm on mobile so I can't see you location. Also try graphite lube. It comes in a small spray can with hose. Last possibility is it broke or a clip came off. Take door panel off and inspect.

 

This is exactly what my 4Runner did when I had the factory SecuriKey alarm disconnected. It is a control module behind the drivers kick panel. My guess is that yours is malfunctioning, or is disconnected.
