checking coil pack resistance

who can tell me if you need to have the coil pack bolted in and connected to a spark plug to test the primary resistance, mine threw a code that no.5 misfired,so I pulled that pack and according to the book(Haynes) they show,(but don't say specifically) testing primary with it in and connected to the plug ,being it's no.5 I can't get the leads in there to test or even see what i'm trying to touch with the probes. so I did it with the coil removed just like they show for the "secondary" test. and no reading,"zeroed out" like touching the probes together, tried it with the next coil over same deal/reading. the "secondary resistance" reading was around 10 for both, I'm not very electrically inclined, so any help would be appreciated, ( 2000 3.4 with original wires and coils 287K) I'm gonna do new wires, do you think I should do all 3 coils as well?
it is doing the very intermittent stuttering under acceleration usually when in low rpms and in overdrive
