There is a diagram under your hood for the vacuum hoses.
Top right goes to the golden thingy at fuel rail. Probably. Other one, can't remember. Length wise, intake plenum. Charcol canister? Your hand is covering stuff.
The other is a water hose.
Tell us what is on the other end of the
hoses.

Yes the fuel pressure regulator needs to be tight, there is likely a torque spec in your FSM but I'm lazy today and not going to dig it up for you, it uses crush washers as a seal to keep the fuel in and needs to hold a seal at ~40 psi.
Your large hose is either the fuel return or the tank vent, push some air thru it if you get bubbling in the fuel tank it's the return line and goes on the fuel pressure regulator otherwise it's the tank vent.
For the vacuum lines trace them back to the origin and consult the vacuum map, or post more pictures.

The torque for the FPR is 22 ft-lbs (you're not going to use a socket on the lock-nut, of course. Either a crows-foot wrench with appropriate offset calculation, or a spring scale on the end of an open-end wrench).
I've never seen an FPR for the 22re (the part number is different than that for a 3VZE), but my FPR uses an o-ring for sealing. Either way, if your FPR lock-nut is loose, either the o-ring or the crush washer is now bad and should be replaced. Don't go cheap here; this is gasoline we're talking about.
