I talked my wife into getting another 4runner the other night and found a good deal on one.
So now I have both a 22re one and a 6cly one both are 2nd gen runners( a 90 and a 95).

I can say the only noticeable differences( at least that i have found so far) between the two are the power, and the engine noise.

The 6cly has a lot more power than the 4cly and is also noticeably more quiet( it doesn't seem to whine like the 4cly does) and I am able to pull out in front of people again and not worry about being rear ended.

I however still prefer my 1990 4cly 22re because of rugged reliability, and the difference in working on the engine.

I just got a 1993 Xtracab SR5 with the 5 speed trans and 3VZE to replace the 1993 Xtracab Deluxe with the 22RE I used to have.

I heard about the problems of the V6 but two friends of mine have the 3VZE engine in their trucks and they works fine. My former 1993 22RE needed a head gasket at about 100K and my other truck with a 22RE with about 160K (257,000 Kms) has a piston noise when cold and low oil pressure...

And unlike 4Runners, SR5 pickups were not available with the 22RE motor in 1993.
