I just purchased a 2001 4runner with only 64k miles on it and everything is in great shape. Brand new tires. The previous owner only used it for weekend outings and drove his BMW during the week. I am really stoked about my find. I have a 95 second gen that I love. The 01 is for my wife, but I may steal it. The only thing that is wrong with it is the rear defrost doesnt work. The previous owner said that the stealer wanted $600 to replace the window to repair the defrost because they told him one of the lead wires was loose. Any thought or ideas about how I might repair this? Anything else that I should know about a third gen? Timing belt has not been done. Am I good till 90K?

I can honestly say I've never used my rear defroster. If you keep the inside of the glass clean it won't fog up. That won't help you if you need it to remove ice though...
Check the fuse first, then the relay and the switch. If it still won't go, check the traces in the glass and see if there are any breaks in them.

yes you are fine on the timing belt until 90K miles. Typically water pump is also done at that time, not probably necessary but everything else is out of the way to do that at same time.

Presuming it to be an automatic-as manual trannys ended in 2000. So good preventative measure when you have the tbelt done is go ahead and do a new radiator to prevent the dreaded "strawberry milkshake"

x2 - may I suggest the Koyo A1998? I replaced mine with the Koyo, excellent fit and quality at a fraction of the OEM price. I buy OEM most of the time but this is a good replacement part. Toyota radiators appear to fail over time rather than after a certain number of miles so just do it with in the next year or so.
