Fuel system problem

I had this same problem a while back but it has been acting great til now. Ok last time when it happened I got in the 4runner and cranked and cranked but nothing would hit. So I got out and took the fuel line off that connects to the regulator on the fuel rail and it started right up. I turned it off and reconnected the line and it started with ease. It came up tonight so I tried 4crawlers idea on jumpering connectors for fp and B+ to see if it was in the electrical system on the fuel pump and no go. So I took the fuel line off again and she cranked right up again. Could this be a regulator problem or maybe vapor locking? Never checked the gas cap to see if it was vapor locked. Any ideas?

Are you talking about the vacuum line connected to the fuel pressure regulator or the return fuel line?

the fuel line that connects to a metal line on the front of the intake and to the regulator.
