4 questions, transfer case, wheel bearing, 3rd member....
1. How much play should there be in the rear axle bearing?
I pulled the axles to get my third member out and there is play in the axle bearing when I wiggle the "brake plate" with it out, but when it is bolted up, there is no play at all when wiggling the tires. All four axles I have are the same amount of play when out of the housing
2. Is it bad if the rear output shaft of the transfer case has a tiny bit of play in it?
I can just barely detect a slight click when I push it up and down from under the truck.
3. How "tight" should the third member be when you spin it by hand?
The 4:10 third that came out of my truck has 112K on it, and is tight with no play, but I can spin it fast by hand and it will continue to spin a half to a full revolution when I let it go.
The 4:30 rear e-locker has much more "drag" and will stop instantly when I let it go. It feels smooth, just more drag.
4. One extra question. Do you guys use the Elocker gaskets, or just RTV only? "and what color RTV please"

When i replaced the rear wheel bearings in my 86 4Runner, the original bearings were so bad that i could wiggle them side to side almost a 1/4 of an inch. The new bearings made a huge diference. Im not sure on the rest of your questions though.
