Misfire in cylinder 5

I have a P0305 code being thrown on my 1999 Tacoma with the 3.4l V6. I've gone through the following steps trying to diagnose the cause:
  • switched the coil with another cylinder, misfire remains on #5.
  • changed the spark plug on #5, misfire remains.
  • checked the fuel injector with a multimeter, reads ~16 ohms.
  • did a compression test, results were as follows:
cylinder 5 - 190psi cylinder 6 - 190 psi

cylinder 3 - 165psi cylinder 4 - 160 psi

cylinder 1 - 190psi cylinder 2 - 175 psi

I also ran some fuel system cleaner through the gas tank just in case there was an issue there, although its been a while and I probably needed to do that anyway.

Now, I feel its important to add that the stuttering doesn't appear until the truck has come to operating temperature and been running for 10 or so minutes. I'm not sure what to make of that.

What is my next step? Do I just need to take it in or is there more I can do before shelling out for a mechanic to check it out?

P0305 means cyl 2 is also suspect. Love how each cylinder has a buddy system through the ignition coil packs.

I would check the plug wire going to 2 and 5 (or replace them all if you're due).
Check the injector at 5...switch with any. Resistance check is one aspect but not a final determinant.
Plug at 2...since shared with the coil pack.

Fair compression readings. How's your valve cover gaskets looking? Man do I have to change mine. I meant head gasket. Tailpipe blowing any coolant?

There can be mechanical misfires too...so things like cam/crank gear or belt teeth going bad.

Or electrical...as in sensors related to the above...though with this you'd prob have problems starting.

Any other issues going on?

So it also just started throwing a P1300 code. Various websites say: P1300 code is for the ignition coil No.1 and is set when there is no IGF signal to the ECM during engine operation.
