Regearing
I am wanting to regear my 00 prerunner(v6 auto) and wanted to know from you guys if i go to 4.56 gears what it will basically harm and improve on my truck. i'm clueless with gearing. i'll be running 265/70/16. any help is appreciated.

most likely you have 4.10. on the little sticker inside your door jamb, there's a code on the sticker which identifies what you have. i can't remember all the details on which code it is, but if you search here for "gear ratio", theres a few threads that tell you which code to read and what it means.
or, call toyota's 800 number w/ your VIN and they'll tell you over the phone.

going to 4.56's from 4.10's is not that much of a jump, you will have slightly better acceleration, but will be turning higher freeway RPM than you would with 4.10's, so your gas mileage may go down, it will not affect the locker at all
