Leakdown test question

I've made a leakdown tester and plan on testing my '97 Tacoma 3RZ-FE. I understand how to test cylinders 1 & 4, but going with TDC, how do you get both the intake and exhaust valves closed at the same time on numbers 2 & 3? Can this be done without taking off the cylinder head cover? Hopefully, it will be as simple as 180° rotation of the crankshaft rather than 360°.

After doing the leakdown test, I discovered 2&3 are 180° from TDC. I put a long narrow screwdriver into the sparkplug opening and watched it rise and fall as I approached that spot.
