The general gist of it is to talk about the one you know something about, hopefully that's your sister. You don't need to worry about talking about the on again/off again nature of their relationship, or really about him at all except for the obligatory "welcome to the family (you weasel)" comment.
I assume you won't be the only, or the first, speaker. I know for my sister it was a small mix of a funny story or two from our childhood, and the "welcome" line. not much more than that.

I worked on my brothers speech for about a week. It was two or three note cards long typed. I had a stomach ache a few weeks before, because I hate speaking in front of 300 people. Maybe it's just me. Anyway, I was nervous a and read it from the cards. After I was done all sorts of people came up to me saying that was the best Best man speech they had heard at a wedding. I'll see if I can't find it. It was very generally speaking. Well I did have a comment about his belly and my balding but, you can tweek it, or just get some ideas from it. To finish my brother was married 4 months. She was one crazy Biatch!!! Why don't people figure this out before hand. I guess like my brother he thought marriage would make things better. Guess what the don't. If your interested let me know, and I'll look for it.

Start it with humor, get a little serious (focus on the positives of your sis and dude), and wrap it up with a nice "we wish you a happy life" - and you could read a verse about love from the Bible (I can help you with that if you are interested).
Spend some time with each of them doing something fun. That will help too.

thanks guys,
im not to nervouse about giving the speech. im a natural speaker, i just havent been to a wedding for 8 years now so not sure how the speech things works, you guys helped theough, edge, thank you, and jsb if you can find it that be cool, if not dont worry bout it. thanks again wade

i have 8 hours before my speech, and i have nothing. any help from anyone? ive got a small start i guess..... i dunno, just not sure what to say about them, so im sure you dont know either! wade

Tell them the good about how you feel about them. make it short and sweet and to the point. It doesn't have to wow anyone just say what you feel.
